No. 54 Enhancement of the Bioavailability of Coenzyme Q10 by γ-cyclodextrin

Background

Our company has an attract attention technology in functional food and personal care fields which raises the water solubility, stability and bioavailability of functional ingredients by using the inclusion property of cyclodextrin (CD).

Recently, coenzyme Q10 (CoQ10) have been focused as a notable anti-aging ingredient. Stable powder of CoQ10 was successfully prepared by inclusion with γ-CD. We also commercialized "CoQ10-γ-CD complex" as functional health food and cosmetics materials and observed the improvements of skin conditions, bone density and knee pain by supplementation of CoQ10-γ-CD complex (* please refer to the newest result of research "4th", "9th" and "14th", of our homepage for details.).

The very low-bioavailability of CoQ10 is supposed to be the cause of its high lipophilicity. So, in this research the bioavailability of CoQ10 was examined in humans using the γ-CD inclusion complex.

Appeal points of CoQ10


Study design


The open-label, single-dose crossover comparative bioavailability trial consisted of two studies (Study 1 and Study 2) was conducted. Healthy 12 males and 12 females were recruited as subjects. A single dose of the test or comparative materials, both containing 30mg of CoQ10 was administered to each subject on Day-1 of each Study. Approximately of 3 mL of whole blood was extracted from each subject at the following collection points : Prior to dosing, 1, 2, 3, 4, 6, 8, 24, 48 hours after dosing. Plasma was separated from collected blood by centrifugation. Plasma CoQ10 concentration was determined by HPLC-ECD. Study 2 was conducted after a 4 week washout period with the same dietary restrictions and fasting condition as Study 1. Subjects were fasted from 21:00 of Day-1 and only water was allowed.

  • CoQ10-MCC (microcrystalline cellulose) mixture : 150mg (containing 30mg of CoQ10) / dose
  • Emulsified-CoQ10 supplement : 300mg (containing 30mg of CoQ10) / dose
  • Water-soluble-CoQ10 supplement : 600mg (containing 30mg of CoQ10) / dose

Pharmacokinetic parameters

Conclusion

CoQ10-γ-CD complex showed significantly higher bioavailability than other CoQ10 supplements due to higher ΔCmax and longer T1/2.

→ CoQ10-γ-CD complex can be useful for the developments of health supplements and drinks with high bioavailability.
