No. 47 Effect of γ-cyclodextrin on absorption of R-α lipoic acid

This research was reported in 27th The Academy of Pharmaceutical Science and Technology (Kobe International Conference Center, Kobe, May. 24~26, 2012).

Background

Our company have the technology which raises the water solubility, stability and bioavailability of a functional ingredient by using the inclusion property of cyclodextrin (CD).

In spite of high anti-aging functions of R-α lipoic acid, its stability and bioavailability are fairly low. So some charming appeal points of R-α lipoic acid can not be shown up easily. Recently, we succeeded the stability of R-α lipoic acid was enhanced dramatically by inclusion with γ-CD (* please refer to the newest result of research "33th" and the "34th" of our homepage for details.), and commercialized "R-α lipoic acid- γ-CD complex" as functional health food and cosmetics materials.

The low-bioavailability of R-α lipoic acid is supposed to be the cause of its low stability. So, in this research the bioavailability of R-α lipoic acid was examined in rats using the stabilized γ-CD complex.

Appeal points of R-α lipoic acid

Evaluation of bioavailability

method

animal : SD rat (8weeks, male, N=4)
Administration : Administrate histamine 1h before 20mg/kg R-α lipoic acid with p.o.
analysis : plasma concentration of R-α lipoic acid were assayed by LC-MS/MS.

γ-CD complexation improved the absorption of R(+)-α lipoic acid

Conclusions

Effect of γ-cyclodextrin on absorption of R-α lipoic acid

γ-CD could enhance not only the stability of R-α lipoic acid, but also bioavailability.

→ R-α lipoic acid -γ-CD complex is prospective as an excellent anti-aging functional ingredient.
